Abstract Indeed human induced pluripotent stem cells
2 b: X. d0 ]5 z; w(hiPSCs) are considered to be powerful tools in regenerative
. {8 _1 T% B$ i+ _+ E7 |$ nmedicine. To enable the use of hiPSCs in the field of
$ v$ B8 a7 Y! J+ q- ?+ ~9 Q  oregenerative medicine, it is necessary to understand the
2 L8 e& U9 i: L+ umechanisms of reprogramming during the transformation2 B+ J2 a* H: o' v
of somatic cells into hiPSCs. Genome-wide epigenetic" A, h- H) Y. ]# T' z1 _3 G- A
modification constitutes a critical event in the generation of2 z0 ^+ ~4 Z$ N, `
iPSCs. In other words, to analyze epigenetic changes in1 J9 C. K+ U* J5 B4 X
iPSCs means to elucidate reprogramming processes. We( N5 @8 l+ B0 Y! K1 t8 ?/ P
have established a large number of hiPSCs derived from

methylation profiles. Comparison analyses indicated that

were similar to those of human embryonic stem cells) s7 n6 Y+ G5 N% ?8 l# Q
(hESCs). However, the profiles of hiPSCs and hESCs" R8 g: ^3 K8 ]$ k3 C/ X
exhibited epigenetic differences, which were caused by
0 p; `: A& Z% A$ a+ I+ c3 `" Lrandom aberrant hypermethylation at early passages.7 |& U( M3 @# x5 b3 ^
Interestingly, continuous passaging of the hiPSCs diminished1 `% y: f8 s8 K- E) a/ M; Z- ~% U
the differences between DNA methylation profiles of- U4 f! f! l: |- \$ M
hiPSCs and hESCs. The number of aberrant DNA methylation. X* l6 u3 P. D( d+ i9 ]5 u) A" ?5 H( ~
