+ B; e, J* `' D6 g3 tDrs.TsaiandMcKayoriginallyidentifiednucleosteminasaproteinabundantlyexpressedinratCNS(centralnervoussystem)stemcellsthatismarkedlydown-regulatedduringdifferentiation,suggestingapossibleroleinstemcellmaintenance.Theresearcherswentontoshowthatnucleosteminisexpressedinvariousadultandembryonicstemcellpopulations,aswellasinsomehumancancercelllines,andthatitsexpressionisconsistentlyturned-offduringthedifferentiationofstemcellsintomorespecializedcelltypes.
/ h5 `# u2 d" d( Z2 w; fUsingtheRNAigenesilencingmethod,Drs.TsaiandMcKaydisruptednormalnucleosteminexpressionpatternsinrodentCNSstemcellsandhumanosteosarcomacancercells.Theyfoundthattheaberrantdown-regulationofnucleostemininthesecellscausedadecreaseincellproliferation,suggestingthattheexpressionofnucleosteminisrequiredforstemcell--andsomecancercell--proliferation.
  L6 ^1 A, W* W2 [& a' m: NAlthoughtheprecisemechanismofnucleosteminactionisnotyetfullyunderstood,theidentificationofagenewhoseproteinproductspecificallypromotestheproliferationofstemcellsandsomecancercellshasimportantclinicalimplicationsforboththeuseofstemcellsinregenerativemedicineaswellasthetreatmentofcancer.AsDr.Tsaiexplains,Thecharacterizationofnucleosteminsuggeststhatauniqueprimitivestateissharedbybothstemcellsandcancercelllines.Theidentificationofcommonmoleculessharedbybothstemcellsandcancercellsmayfacilitatethediscoveryofself-renewingpopulationswithinagiventumorbyevaluatingtheirexpressionlevels.Perhaps,inthefuture,targetingthesecellswillachieveabettertherapeuticoutcome.
