(1)干细胞能够分化为身体上不同细胞类型。就人而言,胚胎干细胞分化为三种细胞胚层---外胚层(ectoderm)、中胚层(mesoderm)和内胚层(endoderm)。但是人们仍然很难清晰地理解某些干细胞为何以及如何分化成特定胚层。不过,正是这些胚层发育为组织和器官。比如,内胚层分化形成胃、结肠和肺,中胚层分化形成血液、骨头和心脏组织,而外胚层则分化形成神经系统(脊椎、周围神经和大脑)、牙釉质、表皮、口腔黏膜、肛门、鼻孔、汗腺、头发和指甲。由此可知,人皮肤细胞与脑细胞、神经细胞相关,因为它们都是外胚层分化形成的。

来自南加州大学凯克医学院、加州橘子郡儿童医院和纽约Good Samaritan医院医学中心的研究人员进行协同研究,成功地通过抑制p53---一个得到广泛研究的抑癌基因编码的一种蛋白---表达将人皮肤细胞转化为脑细胞,这意味着p53突变有助于确定细胞命运---不管是好的还是坏的---而不只是产生癌症的结果。7 v9 V3 c* Z- Y2 d2 M% g

这篇文章我看过,但是我觉得体内任何一种基因都有既定的作用,p53在体内的作用很大,它在抑癌方面确实有作用,其实iPS细胞与肿瘤细胞的关系正在研究中,没有人敢保证iPS以后的应用前景一定是好的,安全的,抑制了p53确实使细胞变得可塑性强了,但是谁又能保证转变成iPS之后不会导致癌症呢?这个前提没有被保证,谁敢应用于临床呢?个人认为干扰体内任何物质的条件下诱导出的iPS细胞都不是安全的~

安全也算是相对的吧~相对得到不治的cancer来说,IPS用于临床也算是一个机会~~

0 Y7 M8 f7 Q" d( T/ t# k相对于iPS,能分化为体内其他任何细胞,因而它们很难控制,是故用于治疗时风险性较大,我觉得这里还是有些区别的,因为抑制p53表达时,人皮肤细胞发展为看起来极其类似于人胚胎干细胞的细胞,只能分化为来自外胚层分化的细胞,即相对而言,细胞分化分化方向较明确一些,但是外胚层则分化形成神经系统(脊椎、周围神经和大脑)、牙釉质、表皮、口腔黏膜、肛门、鼻孔、汗腺、头发和指甲等等,细胞类型还是比较丰富的,如何实现精确和适量控制才是关键。

& Q/ I  Y& C: W0 q! J我认为之所以抑制p53表达后皮肤细胞变得可塑就是因为将p53抑癌基因的检测和修复病变的功能给抑制了,然后就可以随意进行变化了,加上外界条件的刺激,再加上本身的已经导致的不稳定,所以这样就可以导致细胞类型的转变,当然,都是从外胚层分化来的细胞确实有可能有共同的特征,也可能在本质上有某种相似性,但是以抑制p53发生的转变,还是不能解释是因为这些细胞在母源上的相同从而能够发生分化的吧,09年有一篇文章说p53通路抑制iPS的形成,所以抑制p53会提高iPS的效率,恐怕这些是同样的机理吧~  f1 d1 x3 p0 }" A: I+ o& ^7 {
但是这样其实真的不可避免的会产生cancer~所以临床应用还需谨慎啊。。。

In humans, embryonic stem cells differentiate into three families, or germ layers, of cells. The endoderm, for example, leads to formation of the stomach, colon and lungs, while the mesoderm forms blood, bone and heart tissue. In its study, Zhong's team examined human skin cells, which are related to brain and neural cells from the ectoderm.
