Silver et al. (pages 251–261) show that the circadian molecular clock controls the expression and function of the pattern recognition receptor TLR9. In a vaccination model using TLR9 ligand as adjuvant, they demonstrate that mice immunized at the time of enhanced TLR9 responsiveness present weeks later with a substantially improved adaptive immune response. These findings unveil a direct molecular link between the circadian and innate immune systems. This link, which may have important prophylactic and therapeutic implications, is represented on the cover by a “circadian” digital watch indicating TLR9 as the current time.
