MSC脂肪分化后，用油红O染色，但是染色之后该怎么洗涤呢？用水洗还是用异丙醇洗？

应用水洗。用异丙醇可将染料洗下来，可定量检测。: f) P) i6 V7 F9 M8 o

建议用PBS洗，油红O易溶于苯，溶于乙醇和丙酮等。

1.Cell fixation with 4%PFA at RT for 30min.
2. Remove PFA and wash cell with PBS once.
3. Then wash cell with 60% isopropanol for 30 min on a shaker.
4. Remove 60% isopropanol and add working oil red O solution stain for 30 min on a shaker.: | m, {# R- e8 M9 U+ x
5. After 30min remove oil red O stain immediately and wash with PBS for 4 times., M# K! S' x. x# {& Q+ d- R
6. Leave some PBS in the well and take picture.) _2 e. X6 t2 l' m0 B$ F
The working oil red solution should be made fresh from the stock each time.; ?/ B5 D: l- X9 z# m0 v/ @

直接用PBS洗就可以，油红o的储存液是融在异丙醇中的，工作液用蒸馏水稀释

我买的也是gibco的，但是效果不是很好啊! U/ q* z2 e# e6 j7 ]

细胞初始状态很重要；换分化培养基时融合度达到70-80%就可以了，不用等到100%
