请问过夜PFA固定的 EB 如何洗涤残留 PFA，PBS 过夜浸泡洗涤么，残留的 PFA 在 confocal checkup 的时候导致非特异信号影响，谢谢

the negative effect of PFA treatment for overnight is not the remnant PFA in biopsy.  When you treated your biopsy with PFA for overnight, the proteins over-crossliniing by PFA may happen.  Over cross-linking of proteins will elicit auto-fluorescence when you observe your specimen. On the other hand, the antibodies can hardly label the antigens after protein over cross linking in the biopsies.
