C: l: Q* w9 }' p* BRNA干扰是基因沉默进化中保守的过程，在植物中作为自然抵抗外源病毒作用剂。RNA干扰成为生物学中违背遗传规律研究的一个重要工具，RNA干扰在人类基因治疗应用方面有一定潜力，然而有效传递仍是一个重要问题。化学合成的小干扰RNA和短发夹RNA通过转染被引进细胞RNA由RNA聚合酶3转录。siRNA和shRNA用来基因敲除需要一个有效稳定转染活转导过程。这里我们报导横公狗见人细胞稳定表达shRNA慢病毒载体。当未经过药物筛选稳定在人细胞中转导，我们直接通过荧光素没报告基因。证明了慢病毒感染细胞时起到保护作用。我们观察到转导shRNA的细胞基因表达量是相应对照载体的16-4倍。这一模型体系被证明慢病毒载体在稳定表达shRNA中既是细胞基因敲除工具也是在人类细胞被病毒这种外源感染剂时作为抑制剂的一种方法。
