     western 是蛋白印迹，当然用β-actin这种蛋白，而RT-PCR检测的是DNA序列，用的是nat1基因序列，有什么疑问吗？内标只是一个参照。
0 a+ \7 x9 z0 ^6 _& Q% F     RT-PCR中，在不同的PCR反应管中加入已定量的内标和引物，内标用基因工程方法合成。上游引物用荧光标记，下游引物不标记。在模板扩增的同时，内标也被扩增。在PCR产物中，由于内标与靶模板的长度不同，二者的扩增产物可用电泳或高效液相分离开来，分别测定其荧光强度，以内标为对照定量待检测模板。 ' u$ C- D# R" Y! j
     west中，一般内参与要检测的目的蛋白的分子量最好相差5KD以上。选择β-actin肯定是为了更好的分离出其他蛋白的缘故。内参的稳定，是以一个细胞为基本单位，也就是在刺激下，各个细胞的内参量是基本相同，在其他蛋白变化的同时，也不随刺激变化。所以作为参照物。

    minus就是缺失的意思，应该是这么解释。我查看了下原始文献，摘录一段给大家参考（666页左下段）：

No combination of two factors could induce the formation of G418-resistant colonies (Figure 2C). Two combinations of three factors—Oct3/4, Sox2, and c-Myc (minus Klf4) or Klf4, Sox2, and c-Myc (minus Oct3/4)—generated a single, small colony in each case, but these could not be maintained in culture. With the combination of Oct3/4,Klf4, and Sox2 (minus c-Myc), we observed the formation of 36 G418-resistant colonies, which, however, exhibited a flat, non-ES-cell-like morphology. With the combination of Oct3/4, Klf4, and c-Myc (minus Sox2), we observed the formation of 54 G418-resistant colonies, of which we picked 6. Although all 6 clones could be maintained over several passages, the morphology of these cells (iPS-MEF3) differed from that of iPS-MEF4 and iPS-MEF10 cells, with iPS-MEF3 colonies exhibiting rough surfaces (Figure 2D).
