The stochastic and elite models have been proposed for the- I4 P* E& _- F; N
mechanism of induced pluripotent stem (iPS) cell generation. In1 v0 W* X, D7 z t) `( k! t
this study we report a system that supports the elite model. We
2 r9 t5 N, d/ W E% O) M5 jpreviously identified multilineage-differentiating stress-enduring' [# f/ A m3 m4 g- p
(Muse) cells in human dermal fibroblasts that are characterized. n7 f' j; z! t& B
by stress tolerance, expression of pluripotency markers, selfrenewal,
) @8 \0 q" ?% f5 `! Oand the ability to differentiate into endodermal-, mesodermal-,
# Z0 d9 N; F N$ }' a$ i- `and ectodermal-lineage cells from a single cell. They can
4 {) u+ \$ C8 i Y6 ^" G; wbe isolated as stage-specific embryonic antigen-3/CD105 doublepositive) k9 l e: ]' S8 K( ^
cells. When human fibroblasts were separated into Muse
( Z" I! `- U& n$ Z2 qand non-Muse cells and transduced with Oct3/4, Sox2, Klf4, and C- E1 q I7 L4 y% B2 h
c-Myc, iPS cells were generated exclusively from Muse cells but. ~2 Y9 Y6 l: [1 W
not from non-Muse cells. Although some colonies were formed; p- j8 F' I6 Z# S
from non-Muse cells, they were unlike iPS cells. Furthermore, epigenetic; [2 q) G5 p9 @: ?- t( G7 E
alterations were not seen, and some of the major pluripotency
! o+ \ G! B; E/ X( r- T* gmarkers were not expressed for the entire period during
; f- `. P5 m6 X$ k. k+ I% SiPS cell generation. These findings were confirmed further using
\; ], C% O& o6 Ncells transduced with a single polycistronic virus vector encoding
$ M" }* u k* L6 }7 Uall four factors. The results demonstrate that in adult human fibroblasts! t4 Z s9 W/ B" I! F+ I
a subset of preexisting adult stem cells whose properties are
# [+ N" X; B o9 Isimilar in some respects to those of iPS cells selectively become iPS1 |: U4 C" O; n x& ~4 u. d
cells, but the remaining cells make no contribution to the generation0 A: b3 r4 n% N9 N' T, W- C
of iPS cells. Therefore this system seems to fit the elite model4 n" b; u0 G. ~% `8 v
rather than the stochastic model.
