关于细胞污染

最近实验室细胞污染严重，买来的四季青血清中在显微镜下见到8字型透亮的奇怪物质，不会动，有的时候是两个8字型的并在一起。此外，在培养的细胞中看到类似于黑焦虫的小黑点，非常多，有的在小范围内做无规则运动，有的迅速的游来游去，我试过勤换液，离心等方法，但还是无法除去。
6 `2 G9 X( R3 ~9 i/ i9 W/ }我看过很多论坛里说黑焦虫目前是个世界性难题，无法完全除去，只能勤换液控制其不要太多，且不影响细胞活力。那我有疑问，那上海、美国等细胞库的细胞也会有这种所谓的黑焦虫的困扰吗，他们是怎样保证细胞不被污染的呢？, z- U9 g* o1 W8 F5 f: c0 z7 v
大家讨论一下有没有办法去除这样的污染，以及我刚说的8字型的物质是什么。

迅速游来游去的不是黑焦虫，应该是细菌。

污染这么严重，恐怕从试剂到仪器，空气都要重新消毒灭菌才可以再培养了。先用生化培养基查一下污染源在哪里，最好给空气做一个乳酸熏蒸灭菌。现在黄梅天，更容易污染了

我最近还用细胞做了两次colony formation,10天后在显微镜下观察不到贴壁的细胞，也找不到形成的集落，培养液黄色，很澄清，这是为什么，污染吗，不是说这种黑焦虫不影响细胞活力么，郁闷。。。

所谓的黑胶虫，有可能并非生物，而是纤维蛋白凝聚，磷酸钙沉淀，或者是微泡（Endothelial microparticles are small vesicles that are released from endothelial cells and can be found circulating in the blood. The microparticle consists of a plasma membrane surrounding a small amount of cytosol. The membrane of the endothelial microparticle contains receptors and other cell surface molecules which enable the identification of the endothelial origin of the microparticle, and allow it to be distinguished from microparticles from other cells, such as platelets）所以，有可能是你的细胞活力有问题，或者霉菌污染而造成颗粒样物质增多。

不知道大家都用的什么牌子的血清，我们实验室用的四季青新生牛血清，昨天另一家试剂公司来说最近四季青的血清确实存在质量问题，有很多人都反映有黑焦虫的问题，还有没有人用四季青的啊，在400倍境下观察有类似问题吗？
/ z. s" h5 p0 j! E  T9 t我现在在PBS中也加了双抗，冲洗次数增多，还是无法去除黑胶虫。血清暂时还用四季青的，老师不给换，555。。。

  v  l3 \- t* z我们实验室培养皿中各种形态的东西都出现了，8字型都是算常规的。。。& u1 `( g! ?0 m! v

经过严格的环境消毒，更换血清后消失了。
& I( p. B- G- D* U; r9 F4 p+ B& p, m7 D+ Z7 q( s
建议严格将器械消毒，超净台更换初级净化，空气熏蒸。
