For specific restriction enzymes, as PuvI you mentioned here, are sensitive to DNA methylation. For eukaryotes and prokaryotes, there are different DNA methylation system and result in different DNA methylation style. If you want to use PvuI, the concerns are:0 Q2 W6 ?% \; l; N& q
(1) Dam insensitive: the PvuI site in your plasmid, even stocked in Dam+ E.coli can be digested entirely.
0 H* B; s+ B7 w0 u& z* Z5 d(2) CpG sensitive: the PvuI site in eukaryotic genomic DNA cannot be cut by PvuI due to the CpG methylation (except CpG islands)." ^$ D8 Z4 T0 ~
