However, the fragment or cDNA must not contain a polyadenylation signal. The insertion of such sequences between viral LTRs can cause premature cleavage and polyadenylation during transcription of the viral genome. This interferes with the production of viable recombi-nant virions (Coffin et. al, 1997).
