& m6 _5 Z6 E0 N5 `( l( J% xRecent studies have concluded that oncogene-induced "senescence" is a "physiological" mechanism for controlling tumor progression and may be a measure for cancer therapy or even prevention. However, careful evaluation on these reports led to some serious questions on the validity of this cancer therapy. First of all, the strategy may work only for those cancers which really follow the normal to pre-malignant to malignant path. Even so, it is not clear whether the effect will be long-lasting. The introduction of extrinsic oncogenes and their much elevated level of expression are by no means "physiological" and the reported cancer-treatment/prevention effect might just be some artifacts. More importantly, the reported oncogene-induced cellular "senescence" may also occur in normal cells. Thus it may actually cause some harmful side-effects. In fact, it has been reported that the same "senescence"-inducing oncogenic substance or oncogenes were actually involved in the initiation of the pre-malignant mutations and neoplastic formation of "benign" cell mass. Thus, using oncogenic substances or oncogenes to treat cancer will present a conceptual and political problem. Lastly but more importantly, characterizing the "arrest" of "cell cycle" as a phenomenon or consequence of cellular "senescence" is a total misunderstanding. This phenomenon is purely a cell reproduction inhibition which may have no thing to do with cell senescence. Thus, drug researchers and developers should consider all aspects of biology, medicine, and even ethics. A cancer-treatment drug should not just be effective against the cancer cells but also not harmful to the normal cells. |
