Objectives: We reported the first successful use of interferon alpha-2b for treatment of a solid tumor of the mandible in a child. It was noted that the mandible, which was previously resected, regenerated while the patient was on interferon. In subsequent patients, bone formation in mandibular defects has been rapid and exuberant. We hypothesize that interferon induces osteogenic differentiation of stem cells. The purpose of this study is to determine whether mesenchymal stem cells differentiate into bone forming cells under the influence of interferon alpha-2b.9 G( F, B5 ?. ^
Methods: Porcine mesenchymal stem cells (pMSCs) were harvested and isolated from the iliac crest of Yucatan minipigs (age 8 months; n=3). The cells were incubated using standard cell culture conditions (37°C, 5%CO2 , Dulbecco's modified eagle's medium). When culture flasks became near-confluent, cells were detached and re-plated on new culture dishes and the MSCs were grown in the absence or presence of interferon alpha-2b (1,000U/ml; Schering, Lancaster, NY). Experiments were done in triplicate. The cultured cells were monitored microscopically for 21days to evaluate their morphology. On days 3, 7, 14, and 21, cells were stained to detect alkaline phosphatase (ALP). Cell culture plates were also stained with von Kossa to document mineralization.% g* H' t/ K! i7 R6 d+ C7 e; m
Results: Culture dishes incubated with INF alpha-2b demonstrated progressive intensity of staining for ALP positive cells. Whereas culture dishes not receiving IFN showed only a few ALP positive cells. Culture dishes stained with von Kossa demonstrated progressive mineralization whereas those grown in the absence of IFN showed little mineralization.
6 T h) |; d: f Z" GConclusions: The results of this study indicate that IFN alpha-2b induces pMSCs to differentiate into ALP positive cells which appear morphologically similar to osteoblasts and which produce a mineralized extracellular matrix.
