this study we report a system that supports the elite model. We
& r- e3 v. x" m+ F5 c0 Zpreviously identified multilineage-differentiating stress-enduring
( }" h4 w* F" u- m# Z0 C(Muse) cells in human dermal fibroblasts that are characterized
; C1 w9 _1 D T& j. y% m9 z0 hby stress tolerance, expression of pluripotency markers, selfrenewal,2 ]+ F5 x7 l' u, A) ~
and the ability to differentiate into endodermal-, mesodermal-,
# a4 G; z2 {: `. ]; band ectodermal-lineage cells from a single cell. They can
( E$ o4 c/ n2 tbe isolated as stage-specific embryonic antigen-3/CD105 doublepositive: Z2 f( m- q2 [% G
cells. When human fibroblasts were separated into Muse1 u p, W# j0 U& y4 y4 F9 P
and non-Muse cells and transduced with Oct3/4, Sox2, Klf4, and4 T6 }; g' R1 ]2 o8 d, C
c-Myc, iPS cells were generated exclusively from Muse cells but
2 K9 w& U: Z) ~5 Lnot from non-Muse cells. Although some colonies were formed# c1 D/ g; u1 A6 J5 M3 H6 [7 f$ y
from non-Muse cells, they were unlike iPS cells. Furthermore, epigenetic+ ^! R5 V' P6 _% k' E
alterations were not seen, and some of the major pluripotency
9 \' g4 Y. n9 O% n) w8 dmarkers were not expressed for the entire period during
9 J' I- v+ z* Y3 G ziPS cell generation. These findings were confirmed further using
: L k, A: a+ G/ e$ P& o( ccells transduced with a single polycistronic virus vector encoding5 j# d7 K& H+ `2 t& P& k
all four factors. The results demonstrate that in adult human fibroblasts
4 Z; U ?& N5 x1 J$ sa subset of preexisting adult stem cells whose properties are
2 l! ?- H7 L! R5 u. J" Fsimilar in some respects to those of iPS cells selectively become iPS
$ M' i' z! R+ o4 Zcells, but the remaining cells make no contribution to the generation
; T r( J1 p: u0 g: g8 z8 { w+ _of iPS cells. Therefore this system seems to fit the elite model; G# b4 _! _0 n8 Y8 L) ?; I, S$ H# O
