MicroRNA expression profiles classify human cancers (title)

& f' G+ c& z9 e6 E$ z/ `Recent work has revealed the existence of a class of small non-coding RNA species, known as microRNAs (miRNAs), which have critical functions across various biological processes1, 2. Here we use a new, bead-based flow cytometric miRNA expression profiling method to present a systematic expression analysis of 217 mammalian miRNAs from 334 samples, including multiple human cancers. The miRNA profiles are surprisingly informative, reflecting the developmental lineage and differentiation state of the tumours. We observe a general downregulation of miRNAs in tumours compared with normal tissues. Furthermore, we were able to successfully classify poorly differentiated tumours using miRNA expression profiles, whereas messenger RNA profiles were highly inaccurate when applied to the same samples. Yeah, diagnosis is the first step toward therepies. Moreover, miRNA has always been the most prospective choice for gene therapy.

每个mirna的潜在靶点太多了,所以不看好mirna的治疗前景

actually ,the differences should have some relationship with gene trascription and regulation in my opinion ,namely ,the post-translation or the degraduation or the export of mRNA ,all of which can lead to the inconsistency of gene expression and protein ,moreover ,normally a microRNA has many target mRNA ,conversly ,one mRNA also have many sites which can be silenced by microRNA, from this perspective ,it would be not difficult for us to understand the controversies .

Eventhough, one miRNA may have many targetted mRNAs. The possibility is that all these targetted mRNA may work cooperatively rather than separately. This can explain why knocking down one miRNA can induce certain phenotype.
