Although embryonic stem cells currently enjoy the public limelight and show great promise

& ?# s% w9 O5 H; }maladies. The study of mammalian adult stem cells has surged recently, most likely from a
2 d! w% g! L* R# m" S4 k9 V! i6 j! Hmaturation of stem cell studies in the classical developmental model organisms and in
  x  o7 U* O$ _- n1 b' x, G/ [! N4 Yhematopoeisis. All the tissues of the body examined so far are generated and regenerated" p  f/ J0 F1 F9 z
from stem cells, it has been an important first step to adapt or devise new methods to
" \' p. |) R/ M% Eidentify and obtain these cells in quantity and purity for further study. Culture techniques! \: ~) s+ b. F+ E) `0 n
have been optimized for managing the growth and differentiation of stem cells in vitro; as9 D4 p& f% k, y( t6 s+ o
some stem cells are pluripotent, often the method is to guide the fate of such cells among
! p7 o& G8 }% w7 k/ d1 W: \the possible differentiation fates. Much of this work, and that in the classical model organisms,
- @* X' Z" q: L' x& ehas helped define the aspects of the stem cell environment or niche that are crucial: N6 T# \* d2 [: ^* t5 H  q1 P- {6 i
for both growth and differentiation, and these studies have moved in vivo at increasingly
, Z, }9 M5 e9 W4 y6 c( y. O4 Ohigher resolution. Importantly, the in vivo niche is a current target for bioengineering the

Herein, we present methods for studying six types of mammalian stem cells, mammary,1 ^* m; G+ }# ?  j3 S7 _6 D
neural, mesenchymal, endothelial, dendritic, and muscle. We hope the approaches9 B6 p# f  w: T# q2 ~* ^
described in these chapters will assist you in obtaining, characterizing, and studying these) P1 h; n+ C+ W: ^
