for cell based medical therapies, it is the adult stem cells which are responsible for the

maladies. The study of mammalian adult stem cells has surged recently, most likely from a
% Q& [( z. b% L2 |. |( H! zmaturation of stem cell studies in the classical developmental model organisms and in
" j1 D, Y& ^+ `5 W/ Ihematopoeisis. All the tissues of the body examined so far are generated and regenerated: m& c: |: u4 L2 x4 {7 B
from stem cells, it has been an important first step to adapt or devise new methods to
& ^1 D# U0 v, Q# L6 @identify and obtain these cells in quantity and purity for further study. Culture techniques
# g8 O" _/ |; R7 H d: s* dhave been optimized for managing the growth and differentiation of stem cells in vitro; as! P6 n0 i: Z+ Q/ f* {8 I
some stem cells are pluripotent, often the method is to guide the fate of such cells among; q9 V1 @5 d& Y1 h8 W
the possible differentiation fates. Much of this work, and that in the classical model organisms,
, s {/ W; N5 I5 h0 E. phas helped define the aspects of the stem cell environment or niche that are crucial* t' U+ o# z8 c0 V
for both growth and differentiation, and these studies have moved in vivo at increasingly
" i, X7 F8 S- J( V; X4 ~9 `higher resolution. Importantly, the in vivo niche is a current target for bioengineering the
" ?% y9 U4 [; Umatrix and signaling factors.
# ]4 b9 D3 t1 g% x$ ]: W* C6 VHerein, we present methods for studying six types of mammalian stem cells, mammary,, H9 x7 m3 _6 v' K- A* D% |
neural, mesenchymal, endothelial, dendritic, and muscle. We hope the approaches U8 g! q+ R- x, t( F1 |
described in these chapters will assist you in obtaining, characterizing, and studying these# ~+ t2 T" v) N3 l
