for cell based medical therapies, it is the adult stem cells which are responsible for the& }0 |; J- s- F, K2 p4 d7 x
body’s natural ability to fight disease, heal and recover, or fail and succumb to various* C; m* N ~0 |5 `2 O6 Z8 ^- x9 V
maladies. The study of mammalian adult stem cells has surged recently, most likely from a6 m* O+ b# L% N7 _1 s! w
maturation of stem cell studies in the classical developmental model organisms and in4 {; o8 T4 {7 e! z2 @) S* P
hematopoeisis. All the tissues of the body examined so far are generated and regenerated
* ^$ c5 c ^1 ^# h$ H: ^from stem cells, it has been an important first step to adapt or devise new methods to9 E& Y$ g! `: S0 Z
identify and obtain these cells in quantity and purity for further study. Culture techniques
& \! X+ E! T, h% \6 ?) mhave been optimized for managing the growth and differentiation of stem cells in vitro; as" B6 E) b1 F6 o# ?& l0 a2 G
some stem cells are pluripotent, often the method is to guide the fate of such cells among3 z( v" o# k2 [% K, g9 @, ~% }
the possible differentiation fates. Much of this work, and that in the classical model organisms, f- m v; I( ~ G* K$ p
has helped define the aspects of the stem cell environment or niche that are crucial7 q" l! x& v, X" G' f( |, R) s; z
for both growth and differentiation, and these studies have moved in vivo at increasingly! q! q/ |: g, G- F" M& E3 O
higher resolution. Importantly, the in vivo niche is a current target for bioengineering the3 i2 P' Y' I W; |" k( |
matrix and signaling factors.* K: X5 V! K7 F
Herein, we present methods for studying six types of mammalian stem cells, mammary,
( q5 c& o% w* e4 c$ H$ v8 tneural, mesenchymal, endothelial, dendritic, and muscle. We hope the approaches' j" p; S0 j8 Q# {7 K
described in these chapters will assist you in obtaining, characterizing, and studying these% R9 O3 U5 x( L+ V- K, H
cells or for adaptation to your favorite stem cells. One final note, should you find the
