maturation of stem cell studies in the classical developmental model organisms and in
4 k5 E# q' @8 O) A& [7 k: shematopoeisis. All the tissues of the body examined so far are generated and regenerated& o% r6 P/ {- }( F8 n; ~% s6 G4 ?/ [. z
from stem cells, it has been an important first step to adapt or devise new methods to
4 L. f, E& x2 g" T! }0 Aidentify and obtain these cells in quantity and purity for further study. Culture techniques2 k- C+ f% q) O1 Z
have been optimized for managing the growth and differentiation of stem cells in vitro; as4 d5 e/ o; H/ _4 h6 Y' c
some stem cells are pluripotent, often the method is to guide the fate of such cells among* A7 _' {7 u% Z1 n- a: q
the possible differentiation fates. Much of this work, and that in the classical model organisms, {& [) k8 @' {2 F$ _: }0 ^
has helped define the aspects of the stem cell environment or niche that are crucial
: u) Z7 L) t) K0 V& a' Rfor both growth and differentiation, and these studies have moved in vivo at increasingly
- i" b5 O. S+ [( W$ uhigher resolution. Importantly, the in vivo niche is a current target for bioengineering the
9 f9 O* A$ T& w$ rmatrix and signaling factors.
4 z$ d* m" d6 [) t/ ^' PHerein, we present methods for studying six types of mammalian stem cells, mammary,5 x8 j5 ^; v9 ]8 G
neural, mesenchymal, endothelial, dendritic, and muscle. We hope the approaches E# K. @$ J N( T+ P# ]
described in these chapters will assist you in obtaining, characterizing, and studying these
