Although embryonic stem cells currently enjoy the public limelight and show great promise) N1 n, W) V1 j' c0 C/ _) H `
for cell based medical therapies, it is the adult stem cells which are responsible for the
1 A/ V1 `& P7 r7 Y( B+ O) g9 D vbody’s natural ability to fight disease, heal and recover, or fail and succumb to various8 e3 B% b8 h8 w. o. D3 y& o4 `; B8 y
maladies. The study of mammalian adult stem cells has surged recently, most likely from a
6 v4 u2 O) {" b6 u0 X lmaturation of stem cell studies in the classical developmental model organisms and in9 q) D: z8 z) u) q
hematopoeisis. All the tissues of the body examined so far are generated and regenerated: u% N' T6 L6 Y7 V6 @- D
from stem cells, it has been an important first step to adapt or devise new methods to
1 s, Y2 f& e4 Y1 J& O: Bidentify and obtain these cells in quantity and purity for further study. Culture techniques
5 ~& V8 e' q! V0 Ghave been optimized for managing the growth and differentiation of stem cells in vitro; as
1 M$ e: a' Z8 z" X# }some stem cells are pluripotent, often the method is to guide the fate of such cells among
" R6 H, m6 W) H0 ^the possible differentiation fates. Much of this work, and that in the classical model organisms,
& d* V2 a' W2 L3 whas helped define the aspects of the stem cell environment or niche that are crucial0 T& |/ ^ s7 m: E* O
for both growth and differentiation, and these studies have moved in vivo at increasingly) M3 q; H1 y: y' H9 D5 a/ z
higher resolution. Importantly, the in vivo niche is a current target for bioengineering the- \8 u0 Q( X v3 S& T
matrix and signaling factors.
# S ^+ E& c4 L$ z" e# F4 oHerein, we present methods for studying six types of mammalian stem cells, mammary,; y' Q: @: j- \0 L) d0 B8 R, g+ a
neural, mesenchymal, endothelial, dendritic, and muscle. We hope the approaches9 D$ ], s' D; X8 k8 d8 k, u6 [4 J
described in these chapters will assist you in obtaining, characterizing, and studying these5 W* c! d$ C- u0 t: v! [# g& p/ g, b
cells or for adaptation to your favorite stem cells. One final note, should you find the
