Although embryonic stem cells currently enjoy the public limelight and show great promise
+ K, @: o w( u* g0 l5 `( efor cell based medical therapies, it is the adult stem cells which are responsible for the
! I0 b$ E! b! J3 B: t) Dbody’s natural ability to fight disease, heal and recover, or fail and succumb to various& Z& m+ @* b3 d
maladies. The study of mammalian adult stem cells has surged recently, most likely from a
5 l, l/ ~- B7 O: U- s( U% Ematuration of stem cell studies in the classical developmental model organisms and in
' a. @0 b- S8 ]4 I' L! ]hematopoeisis. All the tissues of the body examined so far are generated and regenerated: {. |, x) v: T& {
from stem cells, it has been an important first step to adapt or devise new methods to
% I3 k! g& O E1 r q, Ridentify and obtain these cells in quantity and purity for further study. Culture techniques0 c: r8 l7 K# E1 u4 k" F
have been optimized for managing the growth and differentiation of stem cells in vitro; as- f* e2 D5 n4 W- }9 |9 f2 D
some stem cells are pluripotent, often the method is to guide the fate of such cells among
$ Y- m! S9 }1 Q) [6 o- hthe possible differentiation fates. Much of this work, and that in the classical model organisms,
3 c: P8 [/ T$ ?4 p/ A: ?2 ihas helped define the aspects of the stem cell environment or niche that are crucial6 C# D, }9 f1 k/ M; Q2 i, X
for both growth and differentiation, and these studies have moved in vivo at increasingly% F, r" X6 j& R+ j
higher resolution. Importantly, the in vivo niche is a current target for bioengineering the- u+ @% [: b% ]( W4 h8 j1 O( H
matrix and signaling factors.. x/ P5 i3 D( s6 m4 \. n1 `6 c! n A
Herein, we present methods for studying six types of mammalian stem cells, mammary,
6 w, X- v/ M( U' ?+ H. Z$ zneural, mesenchymal, endothelial, dendritic, and muscle. We hope the approaches
5 r! I$ N( q$ d/ V$ y" c( fdescribed in these chapters will assist you in obtaining, characterizing, and studying these7 Q; @6 R) P6 \
cells or for adaptation to your favorite stem cells. One final note, should you find the/ t9 X7 A/ h+ B" _" ~
