body’s natural ability to fight disease, heal and recover, or fail and succumb to various: b( P# R# w# v! a$ w
maladies. The study of mammalian adult stem cells has surged recently, most likely from a. V6 e8 n* D' M' [4 t
maturation of stem cell studies in the classical developmental model organisms and in5 V4 ], f( t( z& o1 U
hematopoeisis. All the tissues of the body examined so far are generated and regenerated) N: @2 |% X1 h7 X _
from stem cells, it has been an important first step to adapt or devise new methods to
, b/ i6 v" u# f& o! r8 tidentify and obtain these cells in quantity and purity for further study. Culture techniques, R1 y V8 d$ R3 s. S0 ?6 h8 b
have been optimized for managing the growth and differentiation of stem cells in vitro; as
7 g: A9 [7 z1 ~' A! csome stem cells are pluripotent, often the method is to guide the fate of such cells among# h6 G% W6 ]4 w+ m0 e; C( a
the possible differentiation fates. Much of this work, and that in the classical model organisms,: x- c( b( r1 P' O2 V/ C
has helped define the aspects of the stem cell environment or niche that are crucial7 t1 B/ G4 N( m
for both growth and differentiation, and these studies have moved in vivo at increasingly/ E: K$ O1 V5 v, m
higher resolution. Importantly, the in vivo niche is a current target for bioengineering the$ Y) j* p/ y8 E) y) f
matrix and signaling factors.4 R. s6 E) e2 Q$ }: B8 w( u4 W
Herein, we present methods for studying six types of mammalian stem cells, mammary,% n/ p# W" w) ]# H, N
neural, mesenchymal, endothelial, dendritic, and muscle. We hope the approaches
: w+ l' ^1 D' T# B) V9 Zdescribed in these chapters will assist you in obtaining, characterizing, and studying these8 p( B- D& ]' p
