Although embryonic stem cells currently enjoy the public limelight and show great promise
! _9 W0 {( f! t2 ~! rfor cell based medical therapies, it is the adult stem cells which are responsible for the' V1 M& |* X* M/ E9 K
body’s natural ability to fight disease, heal and recover, or fail and succumb to various& T( h0 j) l i ]3 N
maladies. The study of mammalian adult stem cells has surged recently, most likely from a) n$ m8 z8 u) N( @! I
maturation of stem cell studies in the classical developmental model organisms and in
/ v; }# Z+ r+ m* U1 Ghematopoeisis. All the tissues of the body examined so far are generated and regenerated
9 l# S9 [4 h- M* yfrom stem cells, it has been an important first step to adapt or devise new methods to4 l- R: h3 y5 M# @% c* @
identify and obtain these cells in quantity and purity for further study. Culture techniques
( y# x) s9 T; r- Z& l2 whave been optimized for managing the growth and differentiation of stem cells in vitro; as- S7 n$ o. w1 W! J
some stem cells are pluripotent, often the method is to guide the fate of such cells among. |/ d- g$ w" l! F) T5 D
the possible differentiation fates. Much of this work, and that in the classical model organisms,( ]2 S/ F1 f# |9 z! J4 i
has helped define the aspects of the stem cell environment or niche that are crucial
4 h: }, F6 i0 ufor both growth and differentiation, and these studies have moved in vivo at increasingly3 P6 J# D- L$ W. C% s4 n. t9 L7 a6 I
higher resolution. Importantly, the in vivo niche is a current target for bioengineering the$ W8 P. X4 v8 p" @ }: a. k
matrix and signaling factors.
+ T3 c7 n6 Q$ ]* O: s! @* XHerein, we present methods for studying six types of mammalian stem cells, mammary,
1 U& f& P+ A) E; Hneural, mesenchymal, endothelial, dendritic, and muscle. We hope the approaches
: Y& p+ M N2 F3 q% V# edescribed in these chapters will assist you in obtaining, characterizing, and studying these
