( R; v2 L' L$ GItusedtobeconsidereddogmathatanerve,onceinjured,couldneverberepaired.Now,researchershavelearnedthatsomenerves,evennervesinpartsofthebrain,canregenerateorbereplaced.Bystudyingthechemicalsignalsthatencourageorimpedetherepairofnerves,researchersattheUniversityofWashington,theSalkInstitute,andotherinstitutionsmaycontributetoeventualtreatmentsforinjuredspinesanddiseasedretinas,accordingtoapresentationattheannualmeetingoftheAmericanAssociationfortheAdvancementofScience(AAAS).
" O, R5 e( u2 ^, b+ U2 ^( ]Muchofthisresearchfocusesonstemcells,oneofseveraltypesofgeneralcellsthatcangiverisetospecializedcells,likeneurons.Itwasoncethoughtthathumanstemcellswereonlyfoundinembryos,andinbonemarrow,wheretheyproducebloodcells.Butstemcellsarealsobeingfoundinadults,includingthebrainandtheeye.Forexample,stemscellssteadilyreplacedeadneuronsintheolfactorybulb,whichtransmitsscentsignalstothebrain,andthehippocampaldentategyrus,anareathatorganizesshort-termmemory./ O) j; Z/ \5 _% T0 S
However,thepaceofstem-cellrepairsinhumansisslow.Andinsomecases,stemcellscanevenimpedehealing.Stemcellsinaninjuredspinalcordcancreateastickyscarthatblocksnerveregeneration,accordingtoDr.PhilipHorner,anassistantprofessorintheDepartmentofNeurosurgeryintheUWSchoolofMedicine., c& N% k+ A) R# {+ N. }* M
We’vefoundthattheaxons,thepartsofthenervesthattransmitsignals,trytoregenerateafteraninjurybutgetcaughtinthescar.It’slikethey’restuckinthemud,Hornersaid.We’restudyingwaysthatthisprocessisregulatedtoseeifitcanbemanipulatedtopromotehealing.Inotherwords,we’relookingatwaystogettheaxonsoutofthemud.Onewayistomakethemudlessstickybymanipulatingstemcellsthatparticipateinscarformation.Anotheristostimulatetheaxonstopushthroughthescarbyprovidingthecutnerveswithmoleculesthatinduceelongation.We’reusingmolecularsignalscalledgrowthfactorstosimulatethegrowthofculturednervecellsinthelaboratory.

# @0 f/ @/ T( z" MThesametypesofcellsthatcreatescartissueinthespinalcolumncancreatenewcellsintheretinaoftheeye,especiallyinyounganimalsofsomespecies,accordingtoReh.Theretinaisadelicatelight-sensitivemembranethattransmitslightsignalstothebrain.Manyeyesdiseasesthatcauseblindness,suchasglaucomaandasage-related-macular-regeneration,damagetheretina.+ P; k' W# h7 M( P2 Y k) f
Salamandersdon’tgetglaucomabecausetheycanreadilyregenerateretinalcells.Thesameistrueofnewts,frogs,andsometypesoffish.We’retryingtounderstandtheremarkableregenerativepowersoftheselowervertebrates,andthroughthisunderstanding,developstrategiestostimulateregenerationinthehumanretina,Rehsaid.
/ A3 c8 B) c# p& R( J% S) w. }* uWhilesalamanderscanregenerateretinalcellsthroughtheirlife,manyotherspecieslosethisabilityastheyage.Atsomepointineachspecieslifecycle,thestemcellsintheretinamakeatransitionfromaregenerativecelltoacellthatwillmakeascarinresponsetoinjury,likethecellsthatcausescarsinthespinalcord,Rehsaid.Chickensmakethetransitionafewweeksafterhatchinginmostoftheirretina,thoughtheyretainsomelimitedcapacitytoregenerateretinalcellsthroughoutlife.Inrats,it’sonlyamatterofafewdaysafterthecellsaregeneratedthattheylosetheirabilitytoregenerateotherretinalcells.

U/ SHumanretinasseeminglycan’trepairthemselves,yetinrecentstudieshumanretinalcellshavegrownnewneuronswhenculturedinthelaboratory.Thehopeisthatmanyofthemolecularandcellularmechanismsnecessaryforregeneration,thatserveamphibianssowell,arestillinplaceinhumans,Rehsaid.Futurestudiesfromthenervoussystem,aswellasotherorgansystems,shouldenableustodefinetheroadblocksintheregenerativeprocess,anddevelopstrategiestogoaroundthem.& [# \- F6 R L' i) y
