Muchofthisresearchfocusesonstemcells,oneofseveraltypesofgeneralcellsthatcangiverisetospecializedcells,likeneurons.Itwasoncethoughtthathumanstemcellswereonlyfoundinembryos,andinbonemarrow,wheretheyproducebloodcells.Butstemcellsarealsobeingfoundinadults,includingthebrainandtheeye.Forexample,stemscellssteadilyreplacedeadneuronsintheolfactorybulb,whichtransmitsscentsignalstothebrain,andthehippocampaldentategyrus,anareathatorganizesshort-termmemory., M7 n- n$ }" J+ C' z
However,thepaceofstem-cellrepairsinhumansisslow.Andinsomecases,stemcellscanevenimpedehealing.Stemcellsinaninjuredspinalcordcancreateastickyscarthatblocksnerveregeneration,accordingtoDr.PhilipHorner,anassistantprofessorintheDepartmentofNeurosurgeryintheUWSchoolofMedicine." _$ ~ P( B# }$ }- [5 t
We’vefoundthattheaxons,thepartsofthenervesthattransmitsignals,trytoregenerateafteraninjurybutgetcaughtinthescar.It’slikethey’restuckinthemud,Hornersaid.We’restudyingwaysthatthisprocessisregulatedtoseeifitcanbemanipulatedtopromotehealing.Inotherwords,we’relookingatwaystogettheaxonsoutofthemud.Onewayistomakethemudlessstickybymanipulatingstemcellsthatparticipateinscarformation.Anotheristostimulatetheaxonstopushthroughthescarbyprovidingthecutnerveswithmoleculesthatinduceelongation.We’reusingmolecularsignalscalledgrowthfactorstosimulatethegrowthofculturednervecellsinthelaboratory.

Thesametypesofcellsthatcreatescartissueinthespinalcolumncancreatenewcellsintheretinaoftheeye,especiallyinyounganimalsofsomespecies,accordingtoReh.Theretinaisadelicatelight-sensitivemembranethattransmitslightsignalstothebrain.Manyeyesdiseasesthatcauseblindness,suchasglaucomaandasage-related-macular-regeneration,damagetheretina.* A, T) i+ a% q. Salamandersdon’tgetglaucomabecausetheycanreadilyregenerateretinalcells.Thesameistrueofnewts,frogs,andsometypesoffish.We’retryingtounderstandtheremarkableregenerativepowersoftheselowervertebrates,andthroughthisunderstanding,developstrategiestostimulateregenerationinthehumanretina,Rehsaid.! Whilesalamanderscanregenerateretinalcellsthroughtheirlife,manyotherspecieslosethisabilityastheyage.Atsomepointineachspecieslifecycle,thestemcellsintheretinamakeatransitionfromaregenerativecelltoacellthatwillmakeascarinresponsetoinjury,likethecellsthatcausescarsinthespinalcord,Rehsaid.Chickensmakethetransitionafewweeksafterhatchinginmostoftheirretina,thoughtheyretainsomelimitedcapacitytoregenerateretinalcellsthroughoutlife.Inrats,it’sonlyamatterofafewdaysafterthecellsaregeneratedthattheylosetheirabilitytoregenerateotherretinalcells.
( s5 h ]8 K+ A4 s' v0 nHumanretinasseeminglycan’trepairthemselves,yetinrecentstudieshumanretinalcellshavegrownnewneuronswhenculturedinthelaboratory.Thehopeisthatmanyofthemolecularandcellularmechanismsnecessaryforregeneration,thatserveamphibianssowell,arestillinplaceinhumans,Rehsaid.Futurestudiesfromthenervoussystem,aswellasotherorgansystems,shouldenableustodefinetheroadblocksintheregenerativeprocess,anddevelopstrategiestogoaroundthem.
