In modeling community, it is a well known saying that “all models are wrong but some models are useful”. We believe Yamanaka’s stochastic model for generating iPSCs is un-useful because it cannot even be reduced to some basic mathematic descriptions. We further predict that Yamanaka’s stochastic model for generating iPSCs is harmful because it may lead iPS research into a dead end by introducing obscure processes into a system rather than facing the reality head on.

3 ^" R8 c @3 A; n, P' TThe reality is that iPS technique is not a technique for dedifferentiating differentiated normal adult cells back into pluripotent embryonic-like stem cells. It is a powerful technical for non-discriminating oncogenic transformation of any cells including some stem cells 13.

L* uMore specifically, a nuclear genome-integration free and/or genetic engineering-free metabolism mutation-based oncogenesis by Yamanaka factors has been described in details 16. Why not give these explicit hypotheses some tests to see if iPSCs are really induced from (terminally) differentiated cells 17or are just some man-made cancer (stem) cells 13? |
