Depends on which types of colony assay that you are using. If you analyse CFU-C (i.e. Myeloid progenitors), you can distinguish CFU-GEMM, CFU-GM and BFU-E by the color as well (since BFU-E is red/slight red in color due to the presence of hemogloblin). You can take the colonies to make cytospin followed by geimsa staining, by this, you can distinguish the morphology of the colonies as well
