For adult murine stem cells, LSK cells were the basic requirement. However, nowadays, laboratories, which aim at carrying out single-cell transplantation into the mice, use a lot of additional markers to isolate a purer population. E.g CD34 (CD34-), CD150 (CD150+), CD49b (CD49b-) or even adding some Hoechst dye to look for side population (because primitive cells are able to pump out the dye).
! a8 G% W# H$ e1 ] LFor human HSC from Cord blood, there are not so many markers that you can choose from. Typically CD34+CD38-CD90+ contains HSC, but still a hetergeneous propulation. Some labs use Rho123 (more primitive cells are Rho123-/low) or ALDH as well |
