In this issue, Llorca, Lea, Gros, and colleagues reveal the stepwise structural buildup of the pore and host-cell protection from bystander damage. The cover image shows the crystal structure of C5b6, the first step in MAC assembly, and a model for how complement components C6–C9 assemble into an arc based on the cryo-electron microscopy structure of sC5b9.
