(pp. 483–496) describe methods to sequence single neurons from the human brain, enabling study of somatic mutation rates in normal and diseased brains. They perform an analysis of somatic L1 retrotransposition in the cerebral cortex and caudate nucleus and find that somatic insertions are detectable but rare, and they present single-cell genotyping of a pathogenic somatic mutation causing malformation of the cortex. The cover features a single neuron in the shape of a tree. The canopy growing to form the human brain represents the development of the brain during which somatic mutations occur.
