: W5 P$ ?% O9 }组;于术后4周行超声心动图和衄流动力学检查;并处死动物取心脏标本,分别行免疫荧光化学观察移植细胞存
4 N" h; J7 c( o/ y. s活、分化情况及免疫组织化学心肌特异性肌钙蛋白T(cTnT)检测、测定血管密度和胶原容积分数(CVF),计数凋亡
) T l8 ~1 F- R r4 K细胞数。结果犬脂肪组织中含有大量长梭形细胞;术后4周DAPI阳性的ADMSCs可见于梗死边缘区及梗死区,/ x9 r% H; u/ j+ U" t
且分化为心肌细胞表面标志物cTnT阳性的细胞;ADMSCs移植组左室重量指数和左室收缩末容积(LVESV)、非梗( r1 b x0 v ?; Y! U0 ^* B2 X
死区CVF和凋亡细胞数均减少,左室射血分数(LVEF)、左心室收缩压(LVSP)和左心室等容收缩期室内压最大上 _7 e6 ?# X4 e: i( _* ?
升速率(+dp/dtmax)、血管密度和梗死区CVF均增加。结论 同种异体ADMSCs移植后,移植细胞通过分化为心肌
