SCs)进行体外分离培养和4 ,6二脒基一2一苯吲哚盐酸(DAPI)标记,将犬随机分为2组,对照组和ADMSCs移植
' J; T& {: g! M$ o组;于术后4周行超声心动图和衄流动力学检查;并处死动物取心脏标本,分别行免疫荧光化学观察移植细胞存' o" m: H! y9 @! w& M: O! R& Q; _
活、分化情况及免疫组织化学心肌特异性肌钙蛋白T(cTnT)检测、测定血管密度和胶原容积分数(CVF),计数凋亡9 I1 M, e# Q2 M8 e0 ]6 E
细胞数。结果犬脂肪组织中含有大量长梭形细胞;术后4周DAPI阳性的ADMSCs可见于梗死边缘区及梗死区,) Q, A. n: L+ E" }
且分化为心肌细胞表面标志物cTnT阳性的细胞;ADMSCs移植组左室重量指数和左室收缩末容积(LVESV)、非梗2 i0 h0 ?- U" ^+ V6 p7 H$ B9 m
死区CVF和凋亡细胞数均减少,左室射血分数(LVEF)、左心室收缩压(LVSP)和左心室等容收缩期室内压最大上
& ~9 J* V+ X% I5 ^8 |升速率(+dp/dtmax)、血管密度和梗死区CVF均增加。结论 同种异体ADMSCs移植后,移植细胞通过分化为心肌
