SCs)进行体外分离培养和4 ,6二脒基一2一苯吲哚盐酸(DAPI)标记,将犬随机分为2组,对照组和ADMSCs移植
& M* j! C# l7 G' W [+ x6 q( E8 t组;于术后4周行超声心动图和衄流动力学检查;并处死动物取心脏标本,分别行免疫荧光化学观察移植细胞存
/ {" z) e0 a; H/ Y- y活、分化情况及免疫组织化学心肌特异性肌钙蛋白T(cTnT)检测、测定血管密度和胶原容积分数(CVF),计数凋亡
) F5 m7 W2 b5 n* ~0 [. \) m细胞数。结果犬脂肪组织中含有大量长梭形细胞;术后4周DAPI阳性的ADMSCs可见于梗死边缘区及梗死区,: e3 ]" k7 W% H* ^: R
且分化为心肌细胞表面标志物cTnT阳性的细胞;ADMSCs移植组左室重量指数和左室收缩末容积(LVESV)、非梗
( c$ E- b5 R+ u/ N! H5 e' m死区CVF和凋亡细胞数均减少,左室射血分数(LVEF)、左心室收缩压(LVSP)和左心室等容收缩期室内压最大上
