干细胞移植治疗实验犬心肌梗死后心力衰竭的作用.pdf

【摘要】目的初步探讨干细胞移植对犬心肌梗死后心功能的作用。方法取犬脂肪间充质干细胞(ADM—
$ s! g- E, p% ^8 `& X+ MSCs)进行体外分离培养和4 ,6二脒基一2一苯吲哚盐酸(DAPI)标记,将犬随机分为2组,对照组和ADMSCs移植
# U5 I/ g) ]2 U' f组;于术后4周行超声心动图和衄流动力学检查;并处死动物取心脏标本,分别行免疫荧光化学观察移植细胞存
4 H" {% @" Q! _9 P& I7 b6 F活、分化情况及免疫组织化学心肌特异性肌钙蛋白T(cTnT)检测、测定血管密度和胶原容积分数(CVF),计数凋亡
% Z9 a; c+ ^" ?: I细胞数。结果犬脂肪组织中含有大量长梭形细胞;术后4周DAPI阳性的ADMSCs可见于梗死边缘区及梗死区,7 v, m* i# Y [# v( |8 @; I. L
且分化为心肌细胞表面标志物cTnT阳性的细胞;ADMSCs移植组左室重量指数和左室收缩末容积(LVESV)、非梗: n1 I- I3 g' ?: C" J0 q
死区CVF和凋亡细胞数均减少,左室射血分数(LVEF)、左心室收缩压(LVSP)和左心室等容收缩期室内压最大上/ [+ c0 L; O- M5 \% M+ n" M# z
升速率(+dp/dtmax)、血管密度和梗死区CVF均增加。结论 同种异体ADMSCs移植后,移植细胞通过分化为心肌( Q) C$ W$ h4 D! x; U
样细胞,促进新生血管.抑制细胞凋亡,减少非梗死区胶原沉积,改善心肌梗死后的心功能。
