[摘 要]  目的 研究骨髓单个核细胞移植对慢性心力衰竭大鼠心肌的修复作用及其远期效果观察。方法 建
( `$ e. r; ^; o+ K+ b. }5 \0 b- G7 Z立慢性心力衰竭大鼠模型。造模后通过心肌直接注射的方法,于移植组移植DAPI 标记过的骨髓单个核细胞;对照
7 l( {* Q3 z% [, h) I6 u! j组注射同等剂量PBS 溶液;假手术组仅开胸和关胸。并于造模后即刻、移植后4、8 和12 周,分批分组进行免疫组织6 s8 U! c; R$ U/ U1 j0 [' Z
化学和透射电镜的检测。结果 心脏标本切片,荧光显微镜检查证实在细胞移植区有细胞核因被标记而呈荧光的
) X& K3 P/ h( v- U新生心肌和血管。免疫组织化学检查发现细胞移植区有类似心肌细胞的细胞团、心肌特异性结蛋白和肌动蛋白,
1 l9 u8 E% H5 d. o增殖细胞核抗原检测阳性。透射电镜检查可见不成熟的心肌细胞,有细胞超微结构的变化,胞质中有散在肌丝样
8 ^3 i  o1 |* g, o0 ?8 ?物质,还可见内膜不完整的新生血管,且阳性物质随时间推移而逐渐增多。对照组和假手术组则没有这些改变,并; z8 Z9 h; S1 a" D& U5 p
且在不同时间段出现死亡病例。结论 骨髓单个核细胞移植可有效防止慢性心力衰竭大鼠心功能恶化,促进心肌
* Z. F8 U7 A# h和血管再生,在移植后长时间内没有出现新的心肌缺血损害、炎症反应和恶性心律失常,持续修复受损心肌,远期4 H. q7 `6 L  N4 h. z) V# ~

control and sham2operation groups , and at different time , there appeared dead cases.   Conclusions  MBMC transplantation" }2 t7 O6 B0 Y1 t7 ~: p
can effectively prevent heart function deteriorated in chronic heart failure , regenerate cardiomyocyte and blood vessels.  There are, g1 v' M( H* \5 D+ ^
no bad responses such as new ischemia , inflammation and maliganant arrhythmia.  MBMC can continue to repair necrotic myo2
