) n6 |6 v5 X4 _$ U1 ^1 ]% M0 L【中文摘要】 第一章脂肪间充质干细胞的分离、培养和鉴定 目的探讨脂肪间充质干细胞的分离、培养方法，观察其生长增殖情况，鉴定其生物性特征。 方法以S-D大鼠为研究对象，机械分离，胶原酶消化腹股沟和附睾脂肪组织，获取细胞培养于含新生牛血清体积分数为0.1的低糖DMEM培养基中，利用干细胞的附壁生长特性，通过更换培养基而不断纯化获取脂肪间充质干细胞，镜下观察细胞形态变化。MTT法测定传3、5和10代细胞增殖情况。流式细胞仪检测传5代细胞表面CD29、CD34和CD44表达情况。 结果原代培养后24小时看见微少贴壁细胞，初起为小圆形，48小时后可见梭形、短梭形和多角形贴壁细胞，3天后成纤维样细胞明显增多，5天左右进入对数增长期，8天左右细胞单层融合接近90％。传代细胞于2小时即可完成贴壁，生长较快，多数为成纤维样细胞，6天左右细胞单层融合。传3代后细胞形态均一。10代以后细胞则逐渐有宽大梭形样变。生长曲线显示传3、5代脂肪间充质干细胞均有较强的增殖能力，其中传5代细胞更为明显，而传10代细胞的增殖能力逐渐减弱。流式细胞仪检测传5代细胞有95％表达CD44，96％表达CD29，而只有5％表达CD34... 5 Z7 Z% A+ b- T9 T' `0 N# W
