AIM: To study of the effect on the function of rat hepatocytes
  @( O3 J4 q: s5 x- ~cultured with bone marrow cells.
0 F- c5 P3 T$ M% B# J$ y9 ?# yMETHODS: Rat hepatocytes were isolated by the modified) S+ j  Z# F6 ~) |0 y
two-step method described by Seglen. The primary cultured9 T/ l$ E" {! ]+ R) L; ^
hepatocytes and bone marrow cells were served as cocultured0 F# u% l% A) Q7 }1 u  b
group, and single cultured hepatocytes as control group. The. Y' E$ @# i& ]9 J" p* K
yield and viability were assessed by trypan blue exclusion.
  W% F* l! A) l, Z5 h6 gThe morphologic changes of cultured hepatocytes were3 T2 P5 t1 w0 H; N% ]
observed. The concentrations of albumin and urea in the
6 }# c3 m# u; P+ y9 u5 E, Xsupernatant in different cultural period were examined.3 L% R1 O8 g+ K/ R. {- B# \
RESULTS: The albumin synthesis (13.75>2.179, P <0.05)) w! a3 f% x$ J5 u
and urea level (7.27>2.179, P <0.05) had fluctuating
( H2 a6 w/ G% G& y6 U7 Ichanges in one week, and cocultured group had higher. F2 Y, H$ c" y
albumin synthesis and urea level.

养研究骨髓间质干细胞对肝细胞功能的影响以便更好
% ~' e/ w  U  a" `) i( r6 r用于肝细胞移植及生物人工肝.+ L6 I: d) l0 y+ h" q
方法: 采用低浓度胶原酶原位循环灌流法分离大鼠肝细胞% P$ r8 ?: y8 _. a
获得有活性的肝细胞进行原代培养. 台盼蓝排斥法计算细胞
( u" @' E5 b9 ~% m( {$ g  z! _5 l' V产量和细胞活性; 光镜下动态观察细胞形态学改变并收集不同时期培养上清检测其细胞分泌等功能. 比较肝细胞单
4 y$ E& E6 f5 [8 e7 h# r6 ], j纯培养及其与骨髓间质干细胞共同培养时细胞的功能.
9 ?# V# R" D: i( R( X: z' \  c结果: 培养7 d时两组均仍然维持白蛋白分泌及尿素合成功
/ L7 l9 d% ^$ a: k9 i& ^! G' p/ C# G能; 共同培养组与单纯肝细胞培养组在白蛋白分泌 (13.75>
: G+ P: s2 g/ e. Z2.179, P <0.05) 及尿素合成功能 (7.27>2.179, P <0.05) 存在2 e4 a9 e- \3 ?" ~. D8 @$ \* T3 O
显著性差异共同培养组明显高于单纯培养组.
% l% \4 m3 m4 F! s+ e结论: 肝细胞与骨髓间质干细胞共同培养可以使肝细胞- U: G+ h4 B9 ?# ^+ k+ x
维持特异性功能提高肝细胞活性.
