maladies. The study of mammalian adult stem cells has surged recently, most likely from a6 B. Q2 Z8 k/ V: X2 o
maturation of stem cell studies in the classical developmental model organisms and in
q. }/ c9 ]( t% b" P% w: Vhematopoeisis. All the tissues of the body examined so far are generated and regenerated7 M+ ]- a7 ~) o) R) K2 ?: x7 i
from stem cells, it has been an important first step to adapt or devise new methods to
) v/ e- b7 t+ k( u% K7 _ F4 Tidentify and obtain these cells in quantity and purity for further study. Culture techniques& n1 H& P+ Q/ N7 |& T1 _
have been optimized for managing the growth and differentiation of stem cells in vitro; as# E- v' p+ G% T$ {
some stem cells are pluripotent, often the method is to guide the fate of such cells among
! q1 R. H8 W* i7 ~the possible differentiation fates. Much of this work, and that in the classical model organisms,/ {2 b: c0 [1 o! F, r" Z
has helped define the aspects of the stem cell environment or niche that are crucial
4 K; j. i7 q# Z! |' y6 D6 b$ s8 O& ofor both growth and differentiation, and these studies have moved in vivo at increasingly
k; Z( c6 y9 D5 c( f6 w# Y# }- [higher resolution. Importantly, the in vivo niche is a current target for bioengineering the
1 s6 l, \1 U# n/ y$ lmatrix and signaling factors.
; Q B) R1 N5 K2 O: i3 F5 l: \Herein, we present methods for studying six types of mammalian stem cells, mammary,0 }5 E# W/ H! C7 h5 ^
neural, mesenchymal, endothelial, dendritic, and muscle. We hope the approaches+ U* @; F8 b$ H3 K
described in these chapters will assist you in obtaining, characterizing, and studying these
