maladies. The study of mammalian adult stem cells has surged recently, most likely from a8 W% G/ @7 o; N9 H. }
maturation of stem cell studies in the classical developmental model organisms and in
7 v: p! S) {7 _2 \7 h2 v0 Jhematopoeisis. All the tissues of the body examined so far are generated and regenerated: V6 R$ r: `8 d9 c; V: u& W
from stem cells, it has been an important first step to adapt or devise new methods to
- e0 `% o$ }7 E S6 F& p5 o6 Yidentify and obtain these cells in quantity and purity for further study. Culture techniques n# c5 ~* M- s: X* K
have been optimized for managing the growth and differentiation of stem cells in vitro; as
4 p7 m& p O7 U% R# Ssome stem cells are pluripotent, often the method is to guide the fate of such cells among+ p2 C" }% i1 s6 F) Y( G9 B
the possible differentiation fates. Much of this work, and that in the classical model organisms,
9 `) c9 V* w+ N9 Vhas helped define the aspects of the stem cell environment or niche that are crucial7 r6 M4 O" _5 ^* Y! Z/ O
for both growth and differentiation, and these studies have moved in vivo at increasingly/ e7 P& B* G) s$ | b4 p: y
higher resolution. Importantly, the in vivo niche is a current target for bioengineering the3 m: W. T/ M5 D% h
matrix and signaling factors.
6 L' n' I/ X) I" f+ `+ a6 k" [Herein, we present methods for studying six types of mammalian stem cells, mammary,4 H. E! u5 _; l$ R' o: E
neural, mesenchymal, endothelial, dendritic, and muscle. We hope the approaches
% ~8 S, ]; U# i* T- _0 Z& S$ s+ wdescribed in these chapters will assist you in obtaining, characterizing, and studying these% a6 w: J5 v: s {, a) n
