maladies. The study of mammalian adult stem cells has surged recently, most likely from a. A0 ]5 d2 g, P( L# P- j( ]0 |% x
maturation of stem cell studies in the classical developmental model organisms and in: Y- c$ ?- ^6 U9 w
hematopoeisis. All the tissues of the body examined so far are generated and regenerated
5 F- T9 v: v3 D" X/ l; Hfrom stem cells, it has been an important first step to adapt or devise new methods to m$ x1 \0 x9 W, z. }& U8 P
identify and obtain these cells in quantity and purity for further study. Culture techniques
; ?: ], G4 K% n- k& ]- dhave been optimized for managing the growth and differentiation of stem cells in vitro; as t* o! I5 ?) V
some stem cells are pluripotent, often the method is to guide the fate of such cells among
0 h3 n/ C; l9 a! i4 p6 ?" Sthe possible differentiation fates. Much of this work, and that in the classical model organisms,
4 q2 O0 ]4 J o8 `8 Z" Y, Ihas helped define the aspects of the stem cell environment or niche that are crucial4 e7 ` }! q( V" J
for both growth and differentiation, and these studies have moved in vivo at increasingly
- b- K, _8 _. Yhigher resolution. Importantly, the in vivo niche is a current target for bioengineering the
% C; g- W, i! ]7 A/ wmatrix and signaling factors.1 ~- w0 U0 L* E2 T( m1 A
Herein, we present methods for studying six types of mammalian stem cells, mammary,; Y" J: Q5 Q( Y7 ~6 K3 r- n
neural, mesenchymal, endothelial, dendritic, and muscle. We hope the approaches1 B7 s* e* \6 ?% B) ~: Q0 V
described in these chapters will assist you in obtaining, characterizing, and studying these
