关于小鼠骨髓造血干细胞提取和培养

有没有人做过小鼠骨髓造血干细胞提取和培养，能不能请教下你们用的什么方法，用的主要仪器试剂都是从什么公司买的，做一次大概要花多少钱？

分离一般用MACS或者流式，用KSL标记的抗体0 O+ Q* V" t. I7 j1 a) p! A# X
培养的话用无血清培养基，加若干的细胞因子- T8 C* L+ ^- U
你可以到加拿大stemcell公司，他们提供全套的解决方案
& d8 H6 N! U! U* }% }0 p* |和多种培养试剂，还有相应的protocol都很具有参考价值

After isolating femur and tibias from the mouse, obtaining single cell suspension by using mortar and pestle. You can deplet Lineage position population using MACS isolation system, followed by staining the cells by Sca-1, and Kit antibody as Montalexiong mentioned. Lin-Sca+Kit+ contains stem cells but not very pure. If you want to isolate in a higher purity, additionally add CD34 and CD150 antibody as a marker since CD34- and CD150+ populations contains higher frequency of murine stem cells. Antibodies usually are purchased from BD.

我在文献上看到，可以用5’-FU经尾静脉注射来富集造血干祖细胞，目前我也在尝试用这种方法来获取，但是产量很低，用加了细胞因子的培养基培养后，发现有部分细胞变圆变大，剩下的小的细胞活性一直不好。

我们实验室用的是磁珠分选的，一种是磁架分选是美天旎的，一种磁柱分选用的是stemcell公司的，磁珠一般都是三种 lin- 、SCA-1，cd117的，培养基都是stemcell公司的stemspan无血清培养基加三种细胞因子就好了

因子和浓度是：SCF：100ng/ml;IL-3:20ng/ml；Flt-3:100ng/ml
