The stochastic and elite models have been proposed for the

this study we report a system that supports the elite model. We

(Muse) cells in human dermal fibroblasts that are characterized
( p% A$ O$ t# |% ^. M$ Wby stress tolerance, expression of pluripotency markers, selfrenewal,9 s5 }2 J# ]* E
and the ability to differentiate into endodermal-, mesodermal-,
$ B# X, G5 n' c3 x. Yand ectodermal-lineage cells from a single cell. They can* X) g7 r0 Y% f, f/ V3 \/ A3 T
be isolated as stage-specific embryonic antigen-3/CD105 doublepositive& c. w# E2 v% a; [: U0 j0 G. C
cells. When human fibroblasts were separated into Muse' R& u/ s8 D* O! c0 e% q0 J) B2 C
and non-Muse cells and transduced with Oct3/4, Sox2, Klf4, and6 V1 |/ t% Z, h
c-Myc, iPS cells were generated exclusively from Muse cells but0 ~, Q: u) C# L" q; x+ Q! q
not from non-Muse cells. Although some colonies were formed4 T* X: E5 | m V8 H( L1 |
from non-Muse cells, they were unlike iPS cells. Furthermore, epigenetic& N% H, R, w' `# Z
alterations were not seen, and some of the major pluripotency
) P0 e. ?4 B$ G$ Qmarkers were not expressed for the entire period during
4 X) P# Y6 Q& E ^iPS cell generation. These findings were confirmed further using4 v; X# q$ Y- N
cells transduced with a single polycistronic virus vector encoding
! j+ [7 ?( d0 \. t8 \- _$ i3 Oall four factors. The results demonstrate that in adult human fibroblasts
* S# r7 e3 O8 J8 Va subset of preexisting adult stem cells whose properties are
