《Bovine CD49 positive-cell subpopulation remarkably increases in

We previously proved that adult stem cells reside in the bovine mammary gland and possess an intrinsic potential8 W$ e9 d0 I' Q+ Q% D" j/ _$ z
to generate a functional mammary outgrowth. The aim of this study was to investigate on the
T, H: ~5 [7 I: T- c- F# aimmunophenotyping features retained by mammary stem-like cells detected in long term culture. Flow cytometry d; R/ B5 p9 X2 n# s: z
analysis showed different subpopulations of mammary epithelial cells emerging according to the timing of
7 \ c" k# w2 y% ?$ Qcell culture. CD49f+-cells significantly increased during the culture (p b 0.01) and a similar trend was observed,$ T- G( \$ p6 D+ F
even if less regular, for CD29+ and ALDH1 positive cell populations. No difference during the culture was observed
5 f1 k$ L5 h( \- c( R; C3 Ufor CD24 positive cells but after 35 days of culture a subset of cells, CD49f positive, still retained regenerative
7 y& p" r+ @2 `6 ccapabilities in in vivo xenotransplants. These cells were able to form organized pseudo-alveoli when
) Z1 K2 L( h3 U* r. W8 rtransplanted in immunodeficient mice. These results prove the presence of a multipotent cell subpopulation/ P# P+ n: |% x
that retain a strong epithelial induction, confirmed in in vivo xenotransplants with a presumable in vitro expansion3 n2 t8 |0 W ]$ w" u
of the primitive population of adult mammary stem cells. |
