干细胞移植治疗实验犬心肌梗死后心力衰竭的作用.pdf

组;于术后4周行超声心动图和衄流动力学检查;并处死动物取心脏标本,分别行免疫荧光化学观察移植细胞存4 M+ j5 v0 }+ B9 C0 y
活、分化情况及免疫组织化学心肌特异性肌钙蛋白T(cTnT)检测、测定血管密度和胶原容积分数(CVF),计数凋亡
# s) z( h7 m4 q2 [/ {细胞数。结果犬脂肪组织中含有大量长梭形细胞;术后4周DAPI阳性的ADMSCs可见于梗死边缘区及梗死区,+ b0 H" ?6 ?/ L9 {" I# }
且分化为心肌细胞表面标志物cTnT阳性的细胞;ADMSCs移植组左室重量指数和左室收缩末容积(LVESV)、非梗
0 x, c6 e+ p, V9 _) L. p死区CVF和凋亡细胞数均减少,左室射血分数(LVEF)、左心室收缩压(LVSP)和左心室等容收缩期室内压最大上4 M) L8 n5 y* x* b
升速率(+dp/dtmax)、血管密度和梗死区CVF均增加。结论 同种异体ADMSCs移植后,移植细胞通过分化为心肌
